I think actual real demons in DMC would be able to look more human than first generation human/demon hybrids like Dante and Vergil.
Demons like Sparda assume a human form by magic. (That is, of course, assuming that their demonic form is their natural shape, and not, like, a shape that's so habitual it feels natural to them.) Anyhow, real demon demons that naturally look like mutant lizards would have a challenge assuming a human form, and might make many errors, but these errors would be fixable with study and practice. A skilled demon should be capable of appearing indistinguishable from a human, imho.
Hybrids like Dante and Vergil are just walking around in their natural human/demon hybrid bodies, though. Their demon genetics are just, like, there.
Like wolf-dogs, Occicats, Bengals, and Savannah Cats, Dante and Vergil could have traits that Homo sapiens just doesn't have---which is a challenge to dream up, given the wide variety of face shapes, proportions, hair textures, etc. found in humans globally.
We know they have bright white hair, and Dante's appears to have darkened with age. The blue eyes could be bright white Husky dog blue and still function.
Vergil appears to have trouble keeping his hair under control. It's not smooth, it's spiky. That makes sense if the texture is crazy thick, and also he'd have extra trouble if their hair follicles have muscles in them like hackles. Dante's longer hair would hide this better. Nero, if he inherits this feature, could look like a chihuahua trying to kill the mailman when he gets angry now that he's cut it short.
Another cool hair feature that humans don't have, is a transparent cuticle. Best example I can think of is the Bengal cat, or the Satin breed of rabbit. Very glossy, very smooth.
One could posit that demons, hailing from the underworld, have no natural protection from solar radiation. I've seen a hilarious fanfic to this effect, "Devil May Fry."
Larger teeth is an obvious choice. Large teeth means large roots, which in turn means a long face with a heavy jaw: "craggy" features that would make a teenager look older than he is. Maybe creeping toward Looksmaxxer territory.
Those who are into feet have an open playing field. Maybe they only have four toes because of their dad's hooves. Maybe they have opposable big toes and can fold their feet in half lengthwise. Maybe the pointer finger toe is nearly as stout as a human's big toe for better stability while jumping and sprinting.
We could suppose that the reason teenaged Dante was running around shirtless was that any shirt that fit his shoulders looked baggy around his waist and he was too vain to conceal his figure and too lazy to take in the sides.
Their joints are probably very knobby, to anchor powerful muscles. This would be most obvious if they become underweight.
Basically, an average person would know something is weird about Dante, Vergil, and to a lesser extent Nero, but they'd rationalize the oddities because they're obviously People, and only humans are People. That is, of course, assuming the observer had never met another cambion, which most people haven't.
