You ever notice how Givers never have highlights in their eyes, but non-givers do?
Well, I noticed an exception.~
spoilers for season one and the manga up to the latest released chapter!
I fucked up and forgot an important scene! But here's why I don't think it invalidates my theory!

gountess, upon surrendering his vital instrument, has regained the highlights in his eyes, it seems that when a persons vital instrument is given away, or taken away, an individual stops being a giver, gountess's condition may be unique however, as he never really considered the necklace his, as he says, it's rudo's now, he was only holding onto it,
at this point, all givers we see before they gain their powers have highlights in their eyes, and so far, both givers to be shown with them after getting their powers, have been shown either directly before an incident where it is heavily implied her power is taken away, or directly after an instance where the individuals power is given away.
I am convinced that this is an intentional design choice, it doesn't mean every person without highlights is a giver, people consumed by an obsession or intense emotion can temporarily lose their highlights, but we should be paying attention because the eyes tell us a story, wing, the nijiku siblings, hyo, there's more to them than we yet see.
