i think they are unaware. The only time we see them being aware of it is in danganronpa 2 (the reveal of hajime being a reserve course student and the laptop on the 3rd island) and danganronpa 0 and 3 (chiakis relationship with hajime, twilight syndrome and the reserve course secret society (junko is their boss after all)). In danganronpa 0 ryoko is a bit creeped out by the reserve course society, matsuda refers to them as "sheep". In danganronpa 2 fuyuhiko talks about his sister in a rather good way. Mahiru and sato were friends and Nagito has a slight disdain toward the talentless.
