Status: Working On It

Since this is a post and not a dissertation, I will be as brief and concise as I can. I will also keep it as simple as possible so the average reader can understand it. Your life or that of someone you love may depend upon you reading this, passing it on, and helping fund research in this area.

Why is understanding circular DNA so important? 

Simply put, you can’t fix what you don’t understand. Fixing damaged DNA is very important. Normally, our cells do it for us. When they don’t, people can get sick and people can die. 

Currently, science says that each of our chromosomes contains a single linear strand of DNA. They say it like it’s a fact. It couldn’t be further from the truth and yet science works very hard to maintain this myth. Why? My guess is it has something to do with prestige, money, and peer pressure. Isn’t that usually the case?

However, it’s getting more and more difficult for them to prop up this fantasy. They are finding circular DNAs attached to chromosomes and spreading around the cell. DNA circles are able to pass between cells in little messenger packets called exosomes.  Some of these messages may turn out to be Trojan horses, think cancer. 

It’s pretty important to understand how these circular DNAs pop into and out of cells so easily; especially when they are circular instead of “linear” like the chromosome myth states. 

Well, I wish to announce that thanks to A.I., the final nail in the coffin of the linear DNA chromosome model can now be hammered home. I will assume you know little or nothing about biochemistry, so let’s pretend DNA is a very long spiral ladder with two poles and lots of rungs. Both the poles and the rungs are composed of smaller units which under the right chemical conditions can decouple. 

The first step we take is placing a drop of cells onto microscope slides, staining them for DNA, and placing a coverslip on them to spread them out into a thin layer. Photos are taken of the stained living cells.

Next, the slide is briefly dipped into a container filled with dilute hydrochloric acid, and then dipped into another container containing distilled water to rinse off the acid. When they are placed back under the microscope we notice an amazing transformation. The acid releases histone proteins from DNA almost immediately, exposing the circular beaded structures shown below. The chromatin was hiding them. There is no way these circular structures were released from single linear DNA strands. They are far too complex for that.

So what exactly are we looking at here? Well, I have a few ideas even though my future research plans have been thoroughly squelched up to now. I believe that what remains are scaffolding proteins together with DNA. I have plenty of models explaining what these circles are. They may be correct, partially correct, or not correct at all. But for now, it’s all we have until further research is done.

So, you may be wondering, why didn’t I continue this research as a post doc the way everybody does it? Believe me, I tried. My best shot at doing this came after I graduated in 1988. I went down to Chapel Hill, North Carolina and gave a presentation for a post doc position. The results were quite favorable. The lab director told my wife he could get her a job too and to go home and pack up. So we did. Soon, we got a call from Chapel Hill saying that the position had been pulled. No explanation was given and the director would not take my calls. Eventually, I had to go to work for the “man” to pay the bills. My final position at OSU was working for the Pulmonary Division where I generated lots of data for publications. I worked for free on my research as a side gig. That’s when I uncovered the amazing beaded circles shown in the photos above. Then, a grant was unexpectedly pulled right in the middle of my pulmonary work. What papers we got published have been cited but my tenure at OSU was over, including any further lab work on my research. That’s when I took a position at Wright Patterson Air Force base in Dayton, Ohio in 1994. 

I am now 78 and retired. Unless other scientists are willing to step up to the plate, it’s all up to me. If I am able to come up with the proper funding, a local laboratory might be willing to provide a space for me; especially if a graduate student or post-doc was being funded to do this work. If not, I would be willing to do the work without a salary. It shouldn’t take that long to reproduce my work and examine these circles using immunoassays to see if there are indeed any connections between origins of replications, promoters, enhancers, and splice sites. Oops! Finally got technical, didn’t I? Quick summary: those four “things” are short stretches of DNA found throughout the chromosomal DNA. Understanding how they are related and operate together is absolutely essential to understanding how chromosomes work… or do not work.

If you are not a scientist, what can you possibly do to help get this project off the ground? Well, this is where my expertise drops off a cliff. I am no expert at fund raising and have little or no business acumen. That should be obvious by the fact that I was never able to work on this research after I left Ohio State University and moved to Dayton. I still did plenty of research at both places; even undercover research in my former Ph.D. advisors lab (for free). But my paid “day jobs” were doing other people’s research. Got papers published too.

If I can’t raise enough money to fund a spot in a local laboratory, I’ll hand over the funds to the mainstreamers or perhaps to another struggling scientist. Well, here we go again. Something tells me you’re a hired gun. Doesn’t matter to me. You seem to “manufacture evidence” without backing it up with references. I have plenty of references. You obfuscate with vague terminology and fallacious arguments what is clearly in front of everybody’s eyes. Show me evidence that anybody else on the planet has uncovered the structures you see on this post and has a reasonable explanation for them. Go ahead, make stuff up. That’s what you seem to do best!  

I have one more thing to say to whoever is behind this trolling. I find it hard to believe that someone else over the years has never seen the same things as I did under the microscope. The fact that no one else has reported this speaks volumes about the corruption behind our research institutions. Give me a lab pussywillow and I’ll show you how real these things really are!
