Gonna be real, I'm pretty sure Alastor could reasonably know what trans people are (he might have some different terminology given his time period), given he was definitely an adult during the full swing of the Roaring 20s, age of flappers, queers of all stripes and speakeasies among other things, before he died in the 30s.
I agree that he knows different about different genders just not the modern terminology.
A sort of "Oh, is that what they call it these days?" attitude.
