Vinblastine and vincristine are anti-tumor drugs produced by different species of Vinca. Vinca alkaloids depolymerize microtubules and as a result interfere with the normal formation of microtubules during cell division. Extremely low amount of vinca alkaloids in Vinca species and increased demand for chemotherapy are motives to scale up its production by biotechnological approaches. Here, β-tubulin gene family in Catharanthus roseus (vinca rosea) was analyzed to provide enough information for further approaches. 24 β-tubulin coding sequences from plants, mainly Arabidopsis thaliana, were obtained from genebank and aligned to design gene specific primers. Total RNA from leaves of C. roseus were isolated, the cDNA(s) synthesized using specific reverse primers, the amplified fragments cloned in pJET cloning vector and sequenced. Eight clones containing partial β-tubulin cDNAs’ of C. roseus; including an almost full-length coding sequence, were constructed. The comparison of sequencing results with β-tubulin mRNA from Arabidopsis thaliana, Arabidopsis lyrata, Populus trichocarpa, Zea maize, Oryza sativa, Hordeum vulgare, Gossypium hirsutum, Nicotiana attenuate, Solanum tuberosum and lycopersicone sculentum revealed about 85% similarity of nucleotide sequence and about 98% identities at amino acid level. The clustalW alignment analyses underlined a short peptide at position 359-367 with substitutions very specific to C. roseus. Based on our sequencing results, we predicted at least four members of β-tubulin gene family for C. roseus. TUB cDNAs’ from C. roseus sequenced to reveal any differentiation in comparison with non-vinca alkaloid producer plants. It was proposed the sites would carry higher potential for any amino acid alteration providing self-protection in vinca alkaloids producer plants; e.g. C. roseus.

AMD confirms Radeon Instinct MI100 GPU with CDNA architecture for the second half of 2020 AMD is gearing up for a busy holiday. Not only will we have the arrival of new generation consoles with company technology, but also AMD confirms Radeon Instinct MI100 GPU with RDNA 2 architecture, which promises to shake the GPU market in the same proportion as Ryzen processors in the CPU category. The manufacturer should also debut its CDNA architecture, RDNA's "sister" focused on data centers and raw data processing, with the launch of the Radeon Instinct MI100, planned to arrive in the second half of 2020, as pointed out by Mark Papermaster, head of technology from AMD, on your Twitter. AMD confirms Radeon Instinct MI100 GPU - While the RDNA architecture is geared towards gamer GPUs, CDNA was developed for Data Centers, Machine Learning and raw data processing. cDNA Kütüphanesi Nedir?
Bir hücreden izole edilmiş mRNAlardan sentezlenen cDNA kopyaları klonlanarak cDNA kütüphanesi oluşturulabilir. mRNAlar tek iplikli yapılarından dolayı RNAaz enzimleriyle çok çabuk parçalanır. Bu problemi çözmek için mRNA molekülleri DNA karşılıkları olan cDNA’lara(komplementer tamamlayıcı) çevrilir ve bu halde kullanılır.
cDNA Kütüphanesi Nasıl Hazırlanır?
Bir cDNA kütüphanesi mRNAların izole edildiği dönemde aktif olarak genler hakkında bilgi verir. Bu sebeple tıp kütüphaneleri yapılırken çalışılan genin aktif olduğu gelişme dönemi ve hücre tipi dikkate alınmalıdır. cDNAlar olgun mRNAlardan sentezlendikten sonra intron, promatör bölgesi ve regülatör(düzenleyeci) diziler taşımaz. Dolayısıyla bir cDNA klonu, gen yapısı hakkında bir genomik klon kadar bilgi verir.
Bir cDNA kütüphanesi hazırlığında ilk aşama çalışılacak doku ya da hücre tipinden toplam RNAnın elde edilmesi ki o da şu şekilde olur:
mRNAlar toplam RNAlardan 3’uçlarından taşıdıkları poliA kuyrukları sayesinde kolaylıkla ayrılabilirler.
RNA solisyonu, oligo-dT selülöz klonundan geçirilirken poliA kuyruklu RNA’lar oligo-dT ile hidrojen bağlarla bağlanır ve klona tutulur.
mRNA’lar klonun düşük tuz konsantrasyonu bir tamponla yıkanmasıyla elde edilmiş olur.
Bu aşamalardan sonra ters transkriptaz enziminin varlığında mRNA’dan cDNA sentezlenir. Retrovirüslerden izole edilen bir enzim terz transkriptaz RNA’yı kalıp olarak kullanılarak ortamdaki deoksinükleotid 3 fosfatlardan cDNA klonu sentezler. Sentezin başlaması için mRNA kalıbına bağlanacak bir primere ihtiyaç vardır. Bu amaçla oligo-dT primeri ortama eklenir ve poliA kuyruklarına bağlanması sağlanır.
