What's so fascinating about the whole "brother from another lover" mantra is that it's Ruben who comes up with it. Niall doesn't view Ruben as his brother at the start. He's so conflicted in his feelings about Ruben but brotherhood specifically doesn't really seem to enter his mind. And it's understandable why it wouldn't. After all, it's not like Lori and Maura are married thus making them stepbrothers legally. And even if they were, it's doubtful Niall would consider that an "actual" marriage at that stage in his life. But even aside from that, Niall and Ruben hadn't seen each other in years and had no real relationship to base their brotherhood on in his mind.

But Ruben declared them brothers. But not typical brothers. What we do see of Ruben right from the start is he feels something for Niall but he is likely as conflicted as Niall is about those feelings. If not even more so. It's probably easier for Ruben to label these feelings as that of brotherhood. And in some ways that is what they are. But they also aren't. And labelling them as simply "brothers" doesn't feel enough. And what does Ruben know of familial relationships and the feelings he has for Niall? He can't begin to untangle all those feelings. His entire understanding of family and love is built upon what he saw with his father and his mother. Is it any surprise that he can't understand what he feels for Niall?

"Brother from another lover" feels both a way of understanding what he feels while also attaching a safe label to it. And he asks Niall to echo that statement to him. He gets Niall to repeat that statement so Ruben can feel that closeness. And so Ruben can feel that Niall also shares that closeness as he echoes those words to him.
