I looked up the list of TTs from the 1975 tech manual and picked NCC-3830 Messier for the series' hero ship - solely because of the pun potential in the ship's name.

While Messier is a Starfleet ship, the focus is on the medical pod, which is staffed by civilians. Maybe from the Federation equivalent of FEMA? None of the core cast are in Starfleet.

One of the main cast must be a Tellarite, because that is a TOS people who have been criminally underexplored. A female Tellarite, because all of the few Tellarites I can recall ever seeing were always male.

Any other aliens in the main cast have to be obscure ones that have only appeared once or twice, and only from the TOS movies or novels. There can be Vulcans, Andorians etc in the background or supporting cast, but for the main cast, they have to be really obscure. Think Megarite, Arkenite, or Sulamid.

Speaking of underused possibilities, Earth has colonies. Not every Terran has to be from Earth.
