Nung umaga, hinayaan lang kaming gumawa ng thesis sa lab kasi busy din sila.

Nung tanghali naman, nagcheck ulit kami ng results ng 🦀 biochemical test.

Yung methyl red test for e. coli at salmonella: distinct red = positive.

Then sa plate naman na may colonies sa emb at hea, xld, bsa. Kumuha kami ng isolated colony tapos inistreak sa slant.

Bsa, hea, xld for lia at tsia. Lia = violet Tsia = red. Streak sa lia then stab then double stab sa tsia then streak.

Ojt: Wala kami masyadong ginawa kanina. Nagcheck lang ng result sa biochem test ( Brilliant Green Bile Broth for fecal count and EC for total coliform {both may effervescent or bubble sa Durham's tube} [E. coli] ) Yung EC naman inistreak namin sa EMB (dark brown na kami rin yung nagprepare \:D/

Rappaport V. {Dark blue positive:faded blue} for Salmonella ay inistreak naman namin HEA, Xylose lysine d., at Bismuth sulfa(i)te agar. to isolate black colonies.

Then naglipat din kami ng Salmonella at E. coli na galing sa slant sa emb.

Nagprepare ako ng emb:

350 mL distilled = 350 mL sample × 37.5g ÷ 1000L then pour plate stay sa ref

Naglipat ng salmonella at e. Coli control week 3 day 1

May +10 ojt daw for 1 week from visayas

Naglipat ng sample sa rv (blue) at ec (yellowish) at bgbb (green) tapos water bath

Nagcheck ng result sa ymc (pink) [velvety = mold] [10 to 150 valid count] at

biochemical test:

Barret a + b stand 2hrs red = positive

Bumili ng tinapay sa bakery.

Nakalimutan payong, salamin, tsinelas

Nagdissect ng tilapia (black chin)

Test ulit for e. Coli at salmonella

Record yung mga nakaraang test

Half day lang kami ngayon.

Plates:

EMB = Brown (for fecal count) kumuha kami kanina sa plates namin then nilipat sa PCA slant.

XLD = Orange (isolation for E. coli at Salmonella same with HEA at BSA)

HEA = Dark Green

BSA = Matcha

Broth: Kuha sa plate gamit yung butt ng inoculating loop then streak sa TSIA at stab. Then stab 2x yung LIA saka streak. No need to change loop if same plate.

TSIA = Red

BGBB: Brilliant Green Bile Broth (tuloy-tuloy na 48 hrs incubation). Total coliform. Bubbles in durham tube or

LIA = Violet

Then may quiz sa Monday.

Liquids:

Brilliant Green Bile Broth with Durham Tube for the Detection of total coliform. Positive = bubble in durham or effervescent.

Last, pinakita samin ni mam yung result ng yeast and mold na ginawa ni Jason at sir nung Monday.
