Alecnib 150 (Alectinib) is a highly selective and potent ALK and RET tyrosine kinase inhibitor. In nonclinical studies, inhibition of ALK tyrosine kinase activity led to blockage of downstream signaling pathways including STAT 3 and PI3K/AKT and inducedtumor cell death (apoptosis).
Alecnib 150 (Alectinib) demonstrated in vitroand in vivoactivity against mutant forms of the ALK enzyme, including mutations responsible forresistance to crizotinib.The major metabolite of Alecnib 150 (Alectinib) alectinib (M4) has shown similarly in vitropotency and activity.
Based on nonclinical data, alectinib is not a substrate of p-glycoprotein (P-gp) or breast cancer resistance protein (BCRP), which are both efflux transporters in the blood-brain barrier, and is therefore able to distribute into and be retained within the central nervous system. Alectinibinducedtumor regression in nonclinicalmousexenograft models, including antitumor activity in the brain, and prolonged survival inintracranial tumor animal models.

Alecinix 150 mg (Alectinib) is a tyrosine kinase inhibitor that targets ALK and RET. In nonclinical studies, Alecinix (Alectinib) inhibited ALK phosphorylation and ALK-mediated activation of the downstream signaling proteins STAT3 and AKT and decreased tumor cell viability in multiple cell lines harboring ALK fusions, amplifications, or activating mutations. The major active metabolite of Alectinib, M4, showed similar in vitro potency and activity.
Alecinix (Alectinib) and M4 demonstrated in vitro and in vivo activity against multiple mutant forms of the ALK enzyme, including some mutations identified in NSCLC tumors in patients who have progressed on Crizotinib.
In mouse models implanted with tumors carrying ALK fusions, administration of Alecinix (Alectinib) resulted in antitumor activity and prolonged survival, including in mouse models implanted intracranially with ALK-driven tumor cell lines.
