Hughie, who Homelander doesn't even take seriously, is the one who should defeat him because it fits into Hughie's arc (which is to grow up but retain his humanity and not fall into toxic masculinity). Also, from a narrative standpoint, if Homelander fights Billy, Starlight or Ryan it would simply be the narrative rewarding him but if his final fight is against Hughie it means the narrative is denying him that satisfaction as a consequence of his actions.

This would be like Yuji vs Sukuna where Yuji pities Sukuna for how empty his life is compared to the people Sukuna killed and Yuji gives him one last chance to surrender and let him live in exchange for Sukuna releasing Megumi from his control and him returning to his body (Yuji's body). With Hughie and Homelander, something similar could happen, where Hughie shows Homelander how empty he is compared to all those Homelander killed and offers to surrender by fair means (which, like Sukuna, Homelander won't do) or be defeated by foul means, Hughie could kill him but another possible option he can do is... strip Homelander of his powers forever.

I don't know how Hughie could achieve this since it could happen in several ways (using Soldier Boy, using some serum that mimics Soldier Boy's power-removing properties, modifying the anti-supe serum that Billy wants to use so that instead of killing the supers it simply strips them of their powers permanently, etc.) but what I do know is that Hughie has his friends who can help him achieve this. Also, there is no better way to defeat Homelander than to strip him of his powers, that would be a devastating blow to him and even more so if it is Hughie who does it or at least is involved in that plan because we must remember that Homelander has a fragile sense of identity because it only revolves around his powers and condition as a supe and not who he is as a person and if he makes mistakes he does not learn from them because there is always someone who cleans up his mess and no one dares to challenge him because they benefit from it or fear him because of how powerful he is, that is why Homelander never grew as a person and the only thing that gives meaning to his existence and to all the trauma and pain he had to go through is his condition as a supe, this can be seen in the fact that the only clothes we see him wearing are his supe suit.

If Homelander loses his powers at the hands of Hughie it would be the same or worse than dying for Homelander because Homelander without powers is a nobody and if it is Hughie who does it it would be even worse because he was defeated by a normal human that Homelander sees as insignificant and that would completely destroy Homelander because his pride and sense of identity would be shattered so if Hughie does this Homelander could end up going insane (like what happened with Nancy Down from The Craft) or he would beg Hughie to kill him but Hughie would walk away without saying anything and would not kill him but would leave him alive but being a normal human like him.
