Sure thing bro! If you don’t really keep up with the comics then there is a whole side of the relationship you aren’t seeing. They have done quite a few crossovers in the comics and their relationship is just wonderful. Most it is Deadpool making comments such as calling him ‘sweetie’, ‘honey’ or ‘my lover’ which Spiderman doesn’t appear to enjoy but then again, he never stops him. And just small shows of affection and banter between them, mostly from Deadpool but y’know? For example…

They work really well together too when they team up, and they do have a lot of chemistry. There was actually one comic where Spiderman found out about his healing abilities and asked if he felt pain and he said ‘all the time’ or something along those lines. While most wouldn’t worry about hurting Deadpool due to his healing factor, Spiderman always apologized and asked if he was ok if he got injured, and he’s one of the only characters to ever treat Deadpool like that, which really shows Spiderman cares for him, in my opinion anyway.

Also in the back of the Deadpool comics when he answers the fan questions he usually either answers the questions about him and Spiderman, or references to Spiderman in the answers. Some examples…

Another reason (god this turned into an essay) is that Marvel actually ship them! Not only are they giving them this chemistry in the comics, but they made THIS video about their most recent team up. AND one of the artists posted this picture stating that he ships Spideypool:
