Learning to say “STOP”
Let’s be honest, it almost impossible for anyone to really shun negativity and negative people away- they are inevitable and omnipresent. It is sometimes easier to say “ please don’t bring this matter to me as it is affecting me” to people who pass negative comments or share their tormenting experiences. However, what will we do if our mind is that negative voice echoing, haunting and giving us a lugubrious experience?
Personally, I have and am experiencing it. It has never helped me at all. Honestly speaking, I am the kind of person who smiles and ignores the negativity that finds its way towards me. However, when things don’t always go the way you wanted, or the situation is tying your hands hampering your progress, hope is the only way out. What if the hope itself is invisible and is far- distant?
It is natural for us to feel depressed, despaired and dispirited. But we need to find a way out of this. That is life no? How long can be get stuck in that limbo- that we cannot do it, we are stupid, we are useless and do not deserve anything in this world?
Sometimes I wonder why is there so much competition in life if we are all UNIQUE as individuals?

Darkened skies of future - Self-doubt
Experiences in our lives make us who we are today. We are constantly been manipulated, crafted and changed by the situations, learnings and experiences. As much as an empowering experience changes the course of our lives for better, one cannot deny that even the smallest scar can push us into the limbo of self-doubt during tough times.
Self- doubt, a single word that comes with a hefty price to pay! What is self-doubt? Why are we having it? How are we going to come out of it?
What is Self- doubt?
By definition, self-doubt is “ lack of confidence in oneself and one's abilities. “ However, there is more to it. It is the process where we undermine our abilities, our ownself; and in the worst case, we determine that we are useless and there is no point in us existing! Friends, all of us go through self- doubts at some point in our lives.
Why are we having self- doubt?
Even as I write this post, I have so many questions -Am I writing it correctly? Why should I write if no one is reading it? Can I even write well?- especially given my past experience back in school, where my English teacher didn’t believe in my English and thought I cannot even express myself.
You may observe a pattern in my experience. The main reasons for our self-doubts are
Our past intimidating experiences. The mind is easily manipulated and vulnerable to negatives than the positives. Intimidating and demeaning experiences, especially those experienced for a prolonged period of time, can make us believe the negative insights it as true. Over time, we create a mental barrier, that we CANNOT do it!
We are not over it completely. There will always be a point in life where we will have the right people in life and will make our lives better. yet, we may not be able to erase those undermining words from our past negative experience, out of our life. For example, every single time do badly for a vocabulary or a writing test, at the back of my mind, the voice of my English teacher echoes, “ You cannot study even simple English, you cannot pass it”. No matter how many essays or articles I have written since then, all will be masked away from my vision except the voice of my English teacher. That is because I am not over it completely.
HOW TO DO GET OUT OF THIS?
I can hear your internal outcry! There is always some way to get over it.
Journal the negative thoughts. It is important to declutter our minds so that we can have space for positivity to creep in. While journaling, just jot down all the thoughts and emotions. Pour everything out! Scold the person or incident that made you think low of yourself! Let that anger and anguish out!
Talk into the mirror. Self- help is the best help. In front of a full-length mirror, talk to our own reflection. Imagine that our reflection is the reflection of our mind and the inner us. Talk to it aloud, motivate it! Identify past achievements and techniques used to overcome the negative situation. Let our inner mind know and make it realise that we are UNIQUE!
Take a breath; go with the flow: Sometimes it is better to understand that what is meant for us will come and it is better not force things upon ourselves. Do the best to make things better; eventually, it will get better. Don’t expect anything out of the effort, instead, expect yourself give your 100% and think about the results! Time would be the answer to many questions in life!
At any point don’t give upon yourself! Instead give up on the negative experience! SHOUT OUT LOUD AT THE ECHOES IN THE MIND ,“SHUT UP! Don’t control me, I am not dumb!” Get over it! Move on! But Don’t give up on yourself or your abilities.
“ You are braver, stronger, and smarter than you think”
Increasing the Earth’s forests by an area the size of the United States would cut atmospheric carbon dioxide 25 percent.
A simple act can bring a massive change.
Reforestation might be a government initiative, but that should not stop us from making out own initiatives to reduce carbon emissions.
Let's start small.
1. Pot a few plants at your home. All you need is a container, soil, some seeds and water(regularly). It is very low maintenance
2. Choose vegetables and fruits that you often consume or utilise . For example, I have potted tomatoes and aloe vera as I would most probably use them for personal consumption. Go organic!
3. Make sure your plant is growing well and suit your gardening according into your plant's need.
4.Always remember to plant around your house( if you stay in an individual house) or outside your house( if you stay in apartments). Open up your window in the morning to allow the fresh breeze to tour around your house. Trust me you will see a difference. Not only, the smell of the soil calms you mentally, but it also cools down your house with the fresh breeze.
Friends, let's make a concerted effort to make this dream of zero carbon emissions by potting plants at home.
Today's individual effort is tomorrow's community effort!

Reboot your life...
Grappling thoughts...
Humans are just not social beings, but also emotional beings who are very often hurdled by difficult situations. I understand. Under difficult situations, it can be a chore to be grappling with negative emotions and making sure you are still normal and sane. Very often when one is pushed into the wheel of changing emotions, it is very easy for him to be demoralised. Demoralisation, not only exacerbate the status quo; but it can undermine one's self esteem and confidence.
That's when we sink into melancholy and under the extreme situations, we suffer from depression.
For many years, I have been through so many queries about my own self. I have tormenting apprehensions about me not doing well in life; not achieving what I wished for and not being confident about myself. Many at times, I felt aimless and worthless
Often, I was surrounded by more questions mainly on why I didn't I obtain a top score or secure a job that pays me well. This made me question further about my goals and my outlook of what life is. There have been myriad of days I have went doubting my self worth-questioning my existence. Why should I be in a world that doesn't accept me as who I am, rather, expecting me to go by their rule book? Why should I be the first? How can I be master of all trades? What is the need for a random snob to show off his skills and demotivate me? Why should I concur to everything that has been known to work and not try something new? ... The list is never ending...
Ever changing world...
In a fast- paced world, we are, by default, part of a competitive race to make something big out of our lives. Indeed, it once again proves that Darwin was right about his theory about "the survival of the fittest".
However, thanks to increased awareness about stress, suicide and depression. These awareness have lately been a catalyst for a bigger change.
What is truly fascinating is that something that was once a liberty and an asset of original thinkers has now been open to all- being able to choose their own race to run; instead of someone else's, without being judged.
Such awareness have definitely been a turning point of my life.
Over the years, not only have I evolved to be confident, I have learned to be strong mentally and handle tough calls with a smile and maturity.
What did I do different to see a significant change in life?
It's never too late to start again...
Belief- I believed that it is never too late to press the reboot button in life. Life can push us into the moments of strife. But that doesn't mean it is the end. For us to get up and face the battle, we should believe that it is not the end; but the end to a new beginning. Figure out what is the new beginning/ goal.
Redefine the rulebook: Despite being pushed into crowd of the conformists , we should always define our own set of rules. We need to redefine and refine our goals and purpose in life. We should always be accountable for our own life decisions. Take decisions that are one's conscience and righteousness would accept. Ultimately, we benefit and suffer from the results of those decisions. What works for one, may not work for another. So, why not take our own decisions and face the music; rather than dancing to someone else's (conformist) music?
Do mistake with confidence: Friends it is Completely Ok to do mistakes. We should never regret for commiting mistakes or doing something out of our comfort zone. Instead, we should regret for not even challenging ourselves and giving ourselves the chance to do mistakes. Mistakes would nurture us if we have an attitude to learn rather than succeed. Drop the idea of being a perfectionist. A perfect humanbeing is the one with a plethora of imperfections. That's what makes us uniquely human!
Decluttering the mind: From time to time, let's spend at least 10 minutes for ourselves just to clean our minds. A filthy, cluttered room is going to home more insects (and other uninvited visitors). Similarly, a cluttered mind is going to give room to more negativity to brood. On a weekly or monthly basis, let's take time off to introspect to address our inhibitions, fears, times when we possess overconfidence and etc. Overtime, we will start loving and respecting ourselves-which is the first rule of accepting ourselves as whom we are.
Treasure your inner child : no matter how much we grow, never lose the child in us. Children speak their heart; they are constantly curious, creative and inquisitive. They are rebellious, fearless and resilient. Remember, we were once one of them. How difficult is it going to be to take a ride back to our childhood? Let's listen to the child in us and just have some crazy fun!
P. S: This post is purely on my reflections, introspections and experience. This sharing intends mainly to motivate and aspire readers to rediscover their self-worth and benefit from it.

Dosage of Philosophy
Throughout my lifetime, I realised that life is far more dystopic than we have ever envisioned. Thanks to Films and their fantasies. Films fantasise how utopic life can be. It gives one a false thought that life is perfect because it can be "perfect".
It took me all these years to realise, there is no such thing as "perfect". Tbh, being Perfect is one of the worst things. Today, I am glad to have some maturity to distinguish between illusion and reality ( Thanks to those self-help books, Paulo Cohelo, Dr Abdul Kalam, and many others for educating me the reality). Today I am able to understand that failures (imperfections) are one's best friend and not a success (perfections) ; meanwhile, success should be defined by oneself and not by the society he lives in.
After so many months, I truly want to open up my feelings and thoughts that have been racing through my mind. For the past few months, I have been feeling dejected, anguished and rejected as things do not go by my way. Many times, I question my talent and values, which might be the reason for my failures. Thanks to my support system ( family &friends) who still believe in me than me myself. They think a perfect time has yet to come... But honestly, that would be the last idea I would want to buy as well. And my question is when will it be perfect?
Over the years, months and days, I have given up on the word "perfect". It just glorifies the dictionaries and books but not my life.
But I have not given up on two other things. These things have been made, even this flawed indignant phase of my life, a more the "perfect".---Failures and Hope.
Failures are probable and consistent; more consistent than success. Failures are the real superheroes. They bring out the superpower in us. I realise, over the years, the superpower that I have unleashed is patience.
Hope, the best friend of Failure. A four-lettered idea that motivates me to stay alive for that "perfect timing".
Friends, if you had a wonderful day, HOPE to have a better one tomorrow. If your day had been through failure, HOPE to have a better one tomorrow.
Let's accept that failures and hopes are our best friends and own them as ours.As Helen Keller once quoted " When one door closes(failures), opportunity knocks another (hopes)"
HOPE is the panacea to all today's and tomorrows' problems.
Aseptic Techniques- Cell Culture and Maintenance
Culturing and maintaining the cells are the basic techniques of any cell biological experiment. With a few of the following tips, one can surely ensure a contamination-free culture dish.
1. Make sure your plates, pipettes, pipette gun, gloves are sterile. Use ethanol. 70% ethanol is decontaminant.
It is best if you have 2 different labcoats (only if you could afford) for open bench experiments and cell culture. If your lab doesn't have two coats to offer, that is fine. You could use the same coat for both. BUT make sure NEVER to touch your coat with the gloves on that you will be using for cell culture.
A cheaper option is change gloves when doing cell culture. It is best to change gloves if your doing an DNA or RNA extraction OR switching to cell culture experiments. Gloves aren't that expensive anyways.
Do not note that you can reuse the gloves for their respective purposes once you are done with one experiment. Eg. If you have been doing some Westerns. During the blocking, you decided to trypsinise your cells. Then, change gloves before cell culture. While waiting for cells to incubate in trypsin, you may change back to the gloves used for Westerns to continue with it. When it is time to attend to your cells, change back to the gloves for cell culture.
Alright I know it may sound confusing 😕, until you try it. The key idea is to have two separate gloves when you are multi-tasking between open bench and cell culture experiments.
2. Before you step in to put your items into the BSC. Rub your gloves with ethanol. Make sure after that, you do not touch your lab coat or anything that will not go into the BSC.
Spray ethanol on the BSC table/bench. Wipe it clean. If you have an aspirator, check if it is working and clean with ethanol before you put any items inside. You may choose to UV the BSC as well.
Pick up the items one by one. Spray ethanol throughout. Wipe once and spray again before putting the item into BSC.
3. UV: one may choose to UV before or after (or both times) performing cell culture experiments. UV is another layer of protection against any microbial source of contamination.
NEVER UV with: your healthy cells, media containing FBS or any drugs/ reagents.
FBS, drugs or some reagents contain proteins. UV may rupture their structure, causing them to lose their activity.

Vascular vs Alzheimer’s dementia
A common misconception that you would come across often is, dementia is a disease. Friends, let’s get the basics Right!
Alzheimer’s is a disease BUT dementia is a symptom or condition contributing to the disease.
There are four COMMON types of dementia- Alzheimer’s, vascular, frontotemporal and Lewy body dementia. Each have their own distinctive biomarkers and etiology (the cause of a disease). Yet they share a common features/ symptom that classifies them under the umbrella of “Dementia” – the cognitive and executive dysfunction i.e having memory loss, being unable to execute day to day task independently, feeling insecured or experiencing seizures.
Of the four types of dementia, Alzheimer’s dementia is the most common with profound knowledge available on it; followed by the less known and the second most common Vascular dementia. So, how are they similar or different?
Hence, this post will focus primarily give a contrasting perspective between Alzheimer’s dementia ( AD) and Vascular dementia ( VaD)
AD and VaD are often confused to be the same due to their similarities in their symptoms- primarily memory loss, the inability of performing day to day task independently and occasional seizures (Llewellyn et al., 2010). However, the etiology between both the conditions are different.
AD is often associated with an accumulation of amyloid-beta proteins and formation of neurofibrillary Tau protein tangles (Iadecola, 2010). Unlike AD, VaD is a multifactorial and heterogenous vascular disease. It is a consequence of decreased blood flow and deprivation of oxygen and nutrients to the brain due to an acute brain injury or chronic multiple cerebral infarcts (Iadecola, 2010). As a result, an alteration in the neuronal and dendro-synapses lead to the defects in cognition and executive dysfunction (Kalaria, 2018).
VaD is often accompanied by the severe impairments onto more than one cognitive domain (Smith, 2017). It can occur due to sudden brain damage or gradual symptomatic ischemic stroke (Ravona-Springer, Davidson, & Noy, 2003). On the other hand, AD is more likely a gradual process due to lack of physical and mental activity (Ravona-Springer et al., 2003).

Aseptic techniques- as an amateur
Mastering cell culture techniques can be really hard at the start. You have to be extremely cautious over the type of flask you use, the use the pipette in an aseptic manner and the quantity of ethanol to spray (usually profusely and generously, I mean it!). All of this is mainly to achieve one thing- maintain a cell culture without any sort of contamination ( includes yeast, bacterial, any other microbial).
I recall the first time I was learning this, I was cleaning my gloves with ethanol every 5 to 10 minutes -_- ! I have used more ethanol than have consumed water on that day. Even then, it is obvious in any cell biology lab to have consumed ethanol the most than any other reagent!
Like any other skill maintaining a culture was a Herculean task. I had to be extra cautious about my hands being inside the Biosafety Cabinet(figure 1) ALL the time, without my lab coat not touching any of the samples. To be more precise, I was, in fact, measuring how much of hand and lab coat should be exposed in the hood. Mainly because my senior freaked me out about the contamination caused due to non-sterile practices including not using enough ethanol and exposing too much of your lab coat!!!

Then, I had difficulty even holding the pipette gun (figure 2). Juggling between the suck up button and release button made me feel as if my fingers have to grow a few inches longer to manoeuvre between the two buttons. ( Soul crying for help already)

Not to miss the most crucial part which scares you to your death- incubation with trypsin. Trypsin is a reagent which allows the adhered cells to float onto the liquid leaving the adhered surface. Leaving your cells in trypsin too long can cause your cells to die! That would possibly be the worst nightmare you could ever imagine! Unfortunately, you have no idea how long is too long. Finding the sweet spot between too short and too long did take me time to master.
Nonetheless, with more than two years of experience in this field, I could safely say I have mastered the technique now- to an extent whereby now I am ambidextrous when it comes to aseptic technique! :D

Vascular Dementia- what am I?
Affecting one fifth of the world’s population, vascular dementia (VaD) is the second most recurring form of dementia, after Alzheimer’s Dementia (AD). VaD is a severe form of cerebrovascular injury which exacerbates the cognitive and executive function of an individual. Typically, VaD is more prominent in individuals exposed to multiple insults and injury in the brain(Smith, 2017).
VaD was once assumed to be caused by stepwise cerebral infarcts resulting in declination of cognitive and executive function (Rockwood, 2002). However, this definition could be too narrow. The cause of VaD could be due to single or multiple types of vascular lesions (Smith, 2017). Hence, it is associated with a severe form of the vascular cognitive impairment (VCI) (Rockwood, 2002). VCI describes all forms of cognitive dysfunction caused by cerebrovascular diseases.
There are two main clinical forms of VCI. One is caused due to the symptomatic multiple strokes (Smith, 2017). The other is due to the undiagnosed brain impairment which could be identified by either autopsy or imaging of the brain through a magnetic resonance imaging (MRI) or computerised tomography (CT) scan (Rockwood, 2002; Smith, 2017).
In the event of a vascular damage or an ischaemic stroke in VaD, the debris obstructs the blood flow to the brain (Iadecola, 2013). As a result, there is a gradual decrease in blood flow contributing to deprivation of oxygen and nutrients. Lack of oxygen often contributes to a hypoxic condition. Hypoxia promotes oxidative stress, neuroinflammation and neurodegeneration (Iadecola, 2013).
Oxygen and nutrient deprivation are common features of VaD. A review by Allen & Bayraktutan (2009) revealed that the lack of oxygen and nutrients expose the neurons to an elevated levels of lactic acid, causing acidosis. Acidosis increases the concentration of H+ions. H+ ions would stimulate production of the reactive oxidative species (ROS). Accumulation of ROS results in oxidative stress in VaD (Venkat, Chopp, & Chen, 2015). Consequently, the oxidative stress may contribute to neuroinflammation and neuronal damage due to mitochondrial dysfunction (Allen & Bayraktutan, 2009). Besides neuroinflammation, disruption of the blood brain barrier function and seizures are also observed in VaD (Allen & Bayraktutan, 2009; Sen, Capelli, & Husain, 2018)
