Enhanced Production of Polyhydroxyalkanoates from a Bacteria isolated from heavy metal contaminated site using Bio-effluent as a Substrate
PHA accumulating Bacteria was isolated from a Heavy metal contaminated site and identified as Bacillus subtilis by using Biochemical and phylogenetic characterization. The accumulation of PHA in the strain was checked by Sudan Black staining and Fluorescent staining. It was then confirmed using Transmission Electron Microscopy(TEM). Using molasses as the sole source of carbon in the medium, the production of PHA was checked for 48 hours. It was found that the maximum amount of Polymer was produced when 1% Sugar molasses was amended along with Nitrogen sources in the media. The amount of polymer produced was 1.75mg/ml which was comparable to the production of PHA in culture medium used in the laboratory. The PHA thus produced was characterized for ester groups using UV Visible spectrum; CH2 , C=O and CH groups using FTIR, the methyl esters were recognized in GC MS and corresponding carbon and hydrogen skeletons were predicted using NMR.
