Common faults of shaking incubator:

1. No heating: the heating tube is broken or the temperature controller is broken.

2. The whole machine does not work: the power fuse is broken, the power switch is broken or the timer fails.

3. Vibration does not work: Generally, the transmission belt is broken or slipping, and a few are broken because of the debugging controller.

Maintenance of shaking incubator:

1. During continuous operation, the instrument should be checked regularly every three months: check whether water, dirt, etc. have fallen on the motor and control components; clean the dust on the fan; check the fuses, control components and screws. These similar inspections are required in almost all mechanical equipment.

2. Correct use and attention to the maintenance of the instrument to keep it in good working condition can prolong the service life of the instrument.

3. The natural wear and tear of the instrument after long-term use is normal. After the instrument has been used for one year, if abnormal noise is found in the motor, the heating system is abnormal, the transmission part bearings are worn, the heating constant temperature is abnormal, the belt is loose or cracked, and the electronic control component fails, contact the manufacturer in time.

1. Animal cell culture

    Among all cell cultures, the most difficult is animal cell culture. The following  are the special conditions it requires.

    1) Serum:  Animal cell culture often requires serum. The most commonly      used is  calf serum. Serum provides essential growth factors, such as hormones, trace elements, minerals and fats. Here, serum is equivalent to a natural nutrient solution for culture of animal cells.

    2) Support: Most animal cells have the habit of growing adherently. Culture usually uses glass, plastic, etc. as supports.

　3)Gas exchange: The ratio of carbon dioxide and oxygen must be continuously adjusted during the cell culture process to continuously maintain the required gas conditions.

2. Plant cell culture

   1) Light: Plant cells cultured are not very strict with light conditions, because the materials needed for cell growth are mainly supplied by the medium. However, illumination is not only related to photosynthesis, but also related to cell differentiation. For example, photoperiod can regulate sex cell differentiation and flowering. Therefore, light conditions are particularly important in the early plant cell culture process for the purpose of obtaining plants.

  2) Hormones: The division and growth of plant cells require the regulation of plant hormones. Growth-promoting auxins and cell division-promoting mitogens are the most basic hormones. The division, growth, differentiation and individual growth cycle of plant cells are regulated by corresponding hormones.

3. Microbial cell culture

　 Microorganisms are mostly single-celled organisms, and the living conditions in the wild are relatively simple. Therefore, the conditions for artificial culture of microorganisms are much simpler than that of animal and plant cells. Among them, anaerobic microorganism cultivation is more complicated than aerobic microorganisms, because strict anaerobic needs to maintain the concentration of non-oxygen inert gas such as carbon dioxide, while aerobic microorganisms only need to provide sterile oxygen through constant stirring. Microorganisms have less stringent requirements for culture conditions than animal and plant cells.

What does an incubator do for bacteria?

1. Ensure that the nutrients and gas in the liquid medium are even, and the bacteria can grow better.

2. Make the bacteria evenly distributed, and at the same time, ensure the contact between the entire bacteria and the liquid medium to ensure sufficient absorption efficiency.

 Q: “Do I need to invert when cultivating microorganisms in the incubator?”

 A:  It depends on the culture method. If it is a solid plate culture (Petri dish culture), it needs to be inverted.

When the laboratory cultivates microorganisms in an incubator, the general temperature is 35℃-37℃. This is the most commonly used cultivation temperature and is suitable for routine cultivation of most common microorganisms. If it is cultured in a petri dish with solid medium, the petri dish needs to be inverted. There are mainly the following reasons.

1. Slow down the evaporation of water in the culture medium.

2. Prevent condensate drips from affecting microbial growth

3. It can control the spread of colonies and is conducive to the formation of single colonies.

      a.  Prevent ultraviolet light (ultraviolet radiation reduces service life), if you need to use ultraviolet light, you must stick the original protective film on the sticky mat.

      b.  Cleaning the sticky mat: It is recommended to use ultrasonic wave and appropriate detergent to clean it, or clean it with water and allow to dry naturally.

      c.  Do not use cloth or paper towels to wipe.

 1. Biological safety cabinet: 

Biological safety cabinet is a kind of negative pressure equipment, which plays an important role in the new coronavirus nucleic acid detection.  2. Virus inactivation equipment: 

The new coronavirus needs to be inactivated before nucleic acid extraction, so that the high-level structure of the virus protein is destroyed, the protein no longer has physiological activity, and then loses the ability to infect, cause disease, and reproduce, so as to ensure the safety of test personnel. Greatly reduce the risk of inspection personnel being infected. The inactivation method is that the sample is inactivated at a high temperature of 56°C for half an hour in a water bath. Commonly used instruments include steam sterilizers, sterilizers, and water baths.

 3. Nucleic Acid Amplifier: 

Using a nucleic acid amplifier is the last step in nucleic acid detection, and it is also a particularly critical step. PCR technology is equivalent to the natural replication process of DNA, and its specificity depends on the oligonucleotide primers complementary to the two ends of the target sequence, and consists of three basic reaction steps: denaturation-annealing-extension. The inspector puts the amplification system into the amplifier confirms that the amplification program is correct, and then starts. The amplified results are collected by the fluorescence signal collection system in real time, connected to the computer analysis and processing system to obtain a quantitative result output.

 4. Shaker / Incubator shaker:

Pre-treatment of biological samples, used for bacterial culture, fermentation, hybridization, biochemical reactions, enzyme and tissue research that have higher requirements for temperature and oscillation frequency. Laboratory commonly used liquid shakes, microorganisms, bacteria and cell culture.

When you are culturing bacteria, how to choose the best shaking speed ?

Generally, the best speed is determined by the speed of bacterial reproduction.

However, if the secondary metabolites of bacterial reproduction are needed, the best speed can be determined after testing the culture, because the secondary metabolites are not necessarily proportional to the bacterial concentration.

 The high shaking speed is conducive to full dissolution of oxygen and the dispersion of the bacteria, which is mainly to control the shaking speed according to the oxygen demand of the culture bacteria growth and product fermentation.

Some problems about lab instruments will affect your research when you use it . Today, we will talk several common faults and maintenance of light incubator.

1. The compressor of the light incubator is not cooling

a. In the balance mode, the compressor is always in working condition, the cooling effect is better, and the compressor can be effectively maintained.

b. If it is a single-mode work, it will work when the temperature drops to a certain value, and the temperature will fluctuate greatly when the door is opened and closed. Cause the compressor to start frequently, and the start is easy to shorten its service life.

2. The phenomenon of uneven temperature in the light incubator

a. The uneven temperature may be caused by too dense discharge, or because the circulating fan in the incubator does not work.

b. If the light is used for a long time, the bulb will easily heat up, which will also cause uneven temperature.

3. The circulating air vent of the light incubator freezes

Each refrigerated instrument has a defrost cycle during which the compressor does not refrigerate. The ice on the condenser is melted by the heating element. The melted water is likely to gather at the air outlet and cause freezing.

 4. Maintenance of light incubator

a. The light incubator is turned on once or used for a period of time. When the ambient temperature changes, the error between the measured temperature in the working room and the actual temperature must be checked.

b. After each use of the light incubator, the water in the working room must be wiped off and the main power supply of the incubator must be turned off to ensure safety.

c. During normal operation, the load in the box should not be overcrowded, and the air circulation in the box should not be affected to ensure uniform temperature in the box.

d. If the light incubator works continuously for a period of time under refrigeration, the refrigeration effect is not ideal, which may be caused by frosting on the evaporator. At this time, take out the items in the incubator, set the set temperature to 40°C, run it regularly for 200 minutes, and then put it into normal use (usually for 10-15 days of continuous work, the above operation must be performed once).
