Did you just explain dilution to me?
I have a Bachelors in chemistry, and spent ten years doing non-degree-track academic research at an Ivy-league university. I have multiple publications and patents, both in my field and out of it. I decided it was time to get some letters after my name and, after joining a program, was thrilled to find out that I was accepted into my top-choice lab.
Cut to my first week in my new office; I was learning about a new-to-me piece of equipment with biological applications that has a cognate within chemical ones. (For the curious, FPLC was the new one and HPLC is what I know how to use.) My office mates were discussing a new and precious protein they'd acquired, in a very tiny amount, and in a form of dubious purity.
I didn't want to make a novice mistake by suggesting they simply "throw it on the FPLC" as I had no sense of the sample size/concentration required for a run, or what percentage of the protein on hand would be required for such analysis. After asking a few questions that were pointedly about sample size and concentration in relation to the protein on hand, I was treated to the following slow-talk lecture:
"In OUR lab, what we do when we get a protein is we add a liiiiittle BUFFER to it. That way it's in a liquid form, and we can separate it into a BUNCH of little separate tubes, and we can have a small quantity that we want without having to defrost the whole thing! But, since we've added the buffer, you have to remember, it won't be as concentrated and you'll have to do a calculation to find out how much you ACTUALLY have".
One thing that I committed to when I traded ivory towers was that I would be better about sticking up for myself and my knowledge in the face of mansplainers, and i was delighted that I caught this event as it was happening.
"Did you just explain the concept of 'dilution' to me?"
"No, I was just telling you about how we add water to proteins to make it less concentrated so we can use it for different purposes."
"You just explained dilution to me."
