are you attempting to sharpen in photoshop? make sure you are converting to a smart object before running any sharpening commands.

“convert to video timeline” is in the bottom left corner on the timeline panel:

convert for smart filters is in the “filter” menu at the top:  

after this, you should be able to run the smart sharpen command and it will apply to the whole gif.

you shouldn’t need to adjust frame by frame. it should run it for each frame under the same settings all at once. it may still take a good chunk of time to render, but you should be able to walk away and come back to it being complete.

in order to figure out the issue i’d need more information… are you running topaz on a gif that you’ve converted to a timeline and then to a smart object? that’s usually how it’s done… where did you download topaz from, and what version of ps are you using?
