Josuke Higashikata: At this point bisexuality is just a gene in the Joestar's bloodline starting with George II. He's bisexual, I would say gay but he has shown active interest in women in the series

Okuyasu: Gay, I can see the arguments for bi and pan and stuff however I personally see him as a gay man

Koichi: Definitely straight and an ally however I'm just going to put it out there that I think trans-man Koichi is a headcanon to consider

Yukako: Straight 100% and an ally to all except Josuke and Okuyasu she's homophobic to both of them and would maybe be less during pride month (I love her for it though)

Rohan: Gay man.

Yoshikage Kira: labels himself as straight but has repressed heterosexuality and has internalized homophobia although if he didn't have all that bisexual with a preference for women

Yuya Fungami, Akemi, Yoshie, and Reiko: Polyamorous, do I know the organization of their polycule? No I do not all the girls are interested in Yuya how it goes beyond that I don't know
