#like. instead of going hey you didn’t load in enough to assemble relative to the reference genome #it just ran really fast and was like. yeah. dna test came back negative #and I was like ??????#fortuneately it did also generate a summary file and I was like ohhhhhhhh okay I have like. 1% of the reads I need to do this #i think I just didn’t properly download the full files (or the hosting site my coworker used to send them to me didn’t load the full files)#they did download suspiciously fast for read data but it was 1am so I was like. w/e #which is also why I didn’t check the file sizes #to be fair I DID open the fastqs as txt files to check that they looked okay I just didn’t check the length #oh well. easy enough fix assuming the actual files are hosted otherwise I’m just gonna ask my coworker to bring them in on a thumb drive #but yeah it was really funny #I was like dang the assembly was really fast but it was also the first time I specified it could use all of my cores to run #and I was over eager to check the mutation results so I didn’t properly read the summary output first I was like yeah looks like reads #are good quality #I see some number starting with 4 so that’s gotta be 4Mbp covered :)#it was not. it was 400k BASES in the reads #clicks on mutations and am like. uh. hm #that doesn’t seem right.#456 words #lab tag #bioinformatics #also to be fair in the summary stats page while it wasn’t super obvious it did also have a warning that #in case of not enough reads to assemble to reference it will just treat it as no reads

anyway my sage advice as a grad student is. speak up in class. if you don’t understand something, ask. if the professor asks a question, try to answer even if you’re not sure.

you’re allowed to not know things and to be wrong. it’s easy to be worried about looking dumb but pretty much everyone in class either does not care or also has the same question and is glad you asked it. and if you’re answering a question then again they either don’t care or appreciate that you answered it instead of them

there’s no losing. you get things clarified, the prof will remember you were engaged, your classmates will probably be grateful someone else is saying something, and if someone is judging you that’s their problem bc you’re just doing what you need to do to learn? that’s what you’re in school for

#like I’ll caveat that this isn’t always applicable in grade school.#but in college? esp after freshman year?#no one’s gonna be judging you for being wrong or for being a ‘nerd’ and if they do that’s a minority #and that’s their issue #and they’re not gonna make it a problem for you #that’s the only reason I caveat for grade school bc like. grade schoolers are grade schoolers and do stuff like judge and bully people #for being engaged with their education #but college students????? nah. esp not in grad school #456 words #like as long as you’re not asking stuff just to flex you’ll be fine but if you are #then you don’t need this post #also if you’re genuine and you’re worried people will think you are flexing. again probably not. and if someone does?#thats them. bc you’re not

wild how much I am just trusted to do stuff as a grad student?

it’s nice. it’s kinda scary but it’s nice.

#both in lab and in courses and such #like. ye i have a starter experiment to learn specific lab protocols but given my history #my advisor was like okay I’ll be across the hall if you need anything #instead of supervising my every move #she had to go teach and was like ideally I should be back before you have to pour your plates #but we were also both fully aware that when the agar is ready to pour its Ready #and yet she let me pour 50 plates #also wrt to experimental design it was like. hey we talked about some ideas and I read these papers and think maybe this is an avenue?#and she was like. looks good! you can start on that next week :)#and I was like wrow I just get to do my own experimental design based on what I read???? crazy #also courses so far have been ‘here’s the syllabus. you got these projects and due dates. info on what to do is posted. get started’#everyone is also like ‘please come to office hours if you need help’ but #they don’t just hold your hand through everything by default????#like there’s def a huge leap between highschool and undergrad in terms of how much you are treated like an adult #but there’s also a leap between undergrad and grad school #like dang. I’m an adult I guess? I’m trusted to be able to make decisions on how I spend my time #and esp wrt research. what I do #456 words

anyway in case your prescriber never told you, or even if they did:

please do not simply choose to stop taking a medication without first asking a professional how to stop. even if the med isn’t helping or is causing pretty bad side effects. even if you’ve missed doses and felt perfectly fine. you do not know if stopping cold turkey will kill you.

#this is not about people who don’t have a choice in stopping a med #nor is it a place for anecdotes about how you stopped a med cold turkey and instantly felt better #this is about how a lot of medications can be dangerous to suddenly stop taking and how if you have the option #to contact a physician and check how to stop taking the med #you should do so #(also in some cases it may not actually be the med but the dose/format of it. but even assuming you do need to stop)#and yeah not everything will literally kill you but plenty of things will have pretty horrible withdrawal effects #and could still cause permanent harm #and even if there are no withdrawal effects there may be other reasons not to stop a med at the wrong time (ex: antibiotic resistance)#456 words #psa #ask to tag #queue #this post brought to you by yet another person in my life ‘simply deciding to stop taking’ their meds #like I do understand how it can be hard to keep up with meds I really do #and I understand why you might want to stop taking them #I have stopped taking meds and have needed to stop taking meds and it has been helpful #I have also had to stop improperly bc of access reasons #I understand.
